Description

RPS6KB2 encodes a member of the ribosomal S6 kinase family of serine/threonine kinases. This kinase is activated downstream of the mTOR and PI3K signaling pathways and phosphorylates the S6 ribosomal protein, promoting translation and cell growth. It is involved in cell proliferation, survival, and metabolism. Overexpression and mutations have been implicated in various cancers.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer Amplification and overexpression of RPS6KB2 lead to increased mTOR signaling and cell proliferation COSMIC; PMID: 15607980
Prostate cancer Upregulation of RPS6KB2 correlates with poor prognosis and androgen receptor signaling PMID: 22926524
Colorectal cancer RPS6KB2 mutations and copy number gains contribute to tumor growth via S6 phosphorylation COSMIC; PMID: 23535649

Mutation functional classification

Loss of Function (LOF)

Rare missense variants (e.g., p.Arg58Trp) may reduce kinase activity, but no clear LOF phenotype established.

Gain of Function (GOF)

Amplification and overexpression in cancers lead to increased S6 phosphorylation and enhanced mTOR signaling.

Dominant Negative (DN)

No dominant negative mutations reported.

Pathways

• mTOR signaling pathway (KEGG: hsa04150)
• PI3K-Akt signaling pathway (KEGG: hsa04151)
• Insulin signaling pathway (KEGG: hsa04910)

Protein Summary

RPS6KB2 (S6K-beta-2) is a 495-amino acid serine/threonine kinase with an N-terminal kinase domain and a C-terminal regulatory domain. It is activated by phosphorylation at Thr228 and Ser423 via mTORC1 and PDK1. The protein phosphorylates ribosomal protein S6, eIF4B, and other substrates to promote cap-dependent translation and cell growth. It is widely expressed and frequently dysregulated in cancer.
