How Long Does iPSC Gene Editing Take?
The Short Answer
For a well-behaved line and an established protocol:
| Stage | Duration |
|---|---|
| Thaw and expand | 1–2 weeks |
| Edit (RNP or mRNA electroporation) | 1–3 days |
| Selection and recovery | 1–2 weeks |
| Single-cell cloning | 2–3 weeks |
| Clone expansion to sufficient numbers | 2–4 weeks |
| Genotyping, off-target, karyotype | 2–3 weeks |
| Total to validated clones | ~3–4 months |
| Differentiation to a readout (lineage-dependent) | +3–10 weeks |
If knock-in is required rather than knockout, add two to six weeks for donor design and lower efficiency.
What Moves the Timeline
- • Editing format. RNP electroporation is standard; plasmid delivery is slower to clear and more toxic in iPSCs.
- • Efficiency. Low-efficiency edits require more clones screened, which is the largest single variable.
- • Cloning method. Limiting dilution versus sorting changes both recovery and timeline.
- • Karyotype. Abnormal clones must be discarded and the campaign repeated.
- • Line quality. Some iPSC lines grow slowly or differentiate spontaneously, adding weeks.
- • Regulatory documentation. If the line is destined for a regulated use, characterization adds time beyond the research workflow.
Where Projects Lose Time
- Waiting for confluency before editing. Editing works best at defined densities; overgrowth costs a week to recover.
- Screening too few clones. At modest efficiency, screening 24 clones and finding nothing means repeating the whole campaign.
- Skipping early genotyping. Confirming the edit before expanding saves weeks of work on the wrong clones.
- Not planning the assay. Differentiation protocols often need optimization for edited clones; start pilot differentiation in parallel with expansion.
A Realistic Planning Rule
Plan the campaign backwards from the experiment date: phenotype readout requires differentiated cells, which require expanded clones, which require validated genotype. If the project needs the first phenotype in six months, editing should start in month one, not month three.
Need an iPSC editing campaign scoped?
EDITGENE runs iPSC knockout and knock-in campaigns with documented timelines: expansion, RNP editing, single-cell cloning, genotyping, karyotype, and differentiation-competent clone delivery.

Scientific scope and validation note
Timelines depend on line quality, protocol maturity, and edit type; the ranges above are planning estimates for established workflows, not commitments.
Validated clones require genotype, off-target, and karyotype documentation; expansion beyond recommended passage ranges should be avoided.
