How to Validate a Drug Target Before Committing to a Programme
The Short Answer
Four questions, each with a stopping rule:
| Question | Minimum evidence | Stop if |
|---|---|---|
| Is the target causally linked to disease in humans? | Genetic association in the right direction, or functional human data | Genetics contradict the intended mechanism |
| Does perturbing it change the phenotype in cells? | Knockout or knockdown with rescue | No effect at physiological perturbation levels |
| Is it chemically or genetically tractable? | Tool compound, or a viable modality (oligo, degron, cell therapy) | Neither modality is available |
| Does the effect persist in a disease-relevant model? | Primary or iPSC-derived cells, or in vivo | Effect only in immortalized lines |
The third question is often answered last and should be answered early: a validated target with no modality is a biology paper, not a programme.
The Evidence That Moves a Decision
Direction matters more than strength. A strong association with the opposite direction from your intended pharmacology is worse than a weak association in the right direction.
Human genetics is not optional. Targets with genetic support have historically had higher approval success; the absence of genetic support is a risk factor, not a disqualifier.
Perturbation must mimic the drug. An inhibitor reduces activity; a knockout removes protein. Where the mechanism is inhibition, an inducible knockdown or a catalytically dead knock-in is closer evidence than a null allele.
Disease-relevant model before commitment, not after. The most common expensive mistake is a programme that reaches candidate selection with only immortalized-line data.
A Six-Week Package
For a target with existing genetic support:
- Week 1–2: confirm expression and pathway state in two disease-relevant cell models.
- Week 2–4: knockout or inducible knockdown; measure the phenotype; include rescue.
- Week 4–5: dose–response using graded knockdown, to estimate the therapeutic window.
- Week 5–6: chemical or genetic modality feasibility check, including a resistance-mutation strategy if a small molecule is planned.
That package does not prove the target will work; it establishes whether the next twelve months are justified.
Need the cellular package assembled?
EDITGENE builds the evidence for the middle two questions: knockout, inducible knockdown and rescue derivatives, dose–response panels, and disease-relevant models with the readouts that field expects.

Scientific scope and validation note
Target validation reduces risk; it does not predict clinical efficacy. Decisions should weigh the evidence against the modality and the disease.
Associations and cellular phenotypes are context-specific and should be reported with the model and perturbation used.
