Custom Knockout Cell Line Service: Strategy Design, Single-Cell Cloning and Triple Validation
What the Service Covers
Four stages, each with a defined output:
| Stage | What happens | Output |
|---|---|---|
| Strategy design | Exon choice, deletion boundaries, modality, zygosity target | A written strategy with guides and expected allele structures |
| Editing and selection | Delivery optimisation, editing, antibiotic or sorting selection | An edited pool with measured efficiency |
| Single-cell cloning | Cloning, expansion, early genotyping | 2–3 validated candidate clones |
| Triple validation | Junction PCR, Sanger, zygosity, Western blot | The five-item documentation package |
The strategy stage is where most outsourcing fails. A supplier who starts with "send us your gene" has skipped the decision that determines whether the project is two months or six.
What You Receive
| Deliverable | Detail |
|---|---|
| Validated clones | Two independent clones per genotype where the project requires it |
| Genotype | Junction sequence with both breakpoints and primer sequences |
| Zygosity | Homozygous or heterozygous, with the assay used |
| Protein data | Western blot image with loading and positive controls |
| Controls | Parental or non-targeting line from the same campaign |
| Passage record | Passage at freezing and at validation |
| Optional | Transcript-level check; karyotype; off-target panel |
Timeline
| Phase | Duration |
|---|---|
| Strategy and guide design | 1–2 weeks |
| Editing and selection | 2–4 weeks |
| Cloning and expansion | 3–5 weeks |
| Validation and documentation | 2–3 weeks |
| Total | 8–14 weeks |
Difficult loci, essential genes, and primary-like cells move to the upper end or require a modality change.
What We Need From You
- • Gene, transcript, and the exon or region you want disrupted
- • Cell line, with source and passage if you are supplying it
- • Whether you need homozygous or heterozygous clones
- • The intended assay, so validation can be matched to it
- • Any prior guide or delivery data, including failures
Honest Limits
- • Essential genes cannot be delivered as constitutive knockouts. We will propose inducible knockdown or degron lines instead.
- • Primary cells usually cannot be cloned; the deliverable becomes a validated pool.
- • Efficiency is locus- and cell-type-specific. We report pool-level editing before cloning, and advise whether the clone number is realistic.
- • We do not guarantee a phenotype. We guarantee the genotype, the protein data and the documentation.
Frequently Asked
Can you work with our line?Yes, with a receipt QC and a pilot edit. If the line proves uneditable, the pilot report explains why and what we recommend instead.
Do you provide the guides?Yes, with sequences and a design rationale, so the work is reproducible in your hands.
What if only one clone validates? We continue until two validate or the campaign is exhausted; if the latter, we report the allele spectrum and recommend a strategy change rather than delivering a single clone quietly.
Start With the Strategy
Send the gene, the cell type and the intended assay. The first output is a written strategy — before any editing is scheduled.
Contact EDITGENE to request a project consultation or quote.
Scientific scope and validation note
Timelines are planning ranges for established workflows and depend on locus, cell type, and delivery feasibility.
Validation confirms clones at the passage tested; documentation is provided so that results can be reproduced and audited.
