| Summary |
This gene encodes a serum protein found in association with the major histocompatibility complex (MHC) class I heavy chain on the surface of nearly all nucleated cells. The protein has a predominantly beta-pleated sheet structure that can form amyloid fibrils in some pathological conditions. The encoded antimicrobial protein displays antibacterial activity in amniotic fluid. A mutation in this gene has been shown to result in hypercatabolic hypoproteinemia.[provided by RefSeq, Aug 2014]

FAQ
Which is better for studying B2M function, B2M Knockout A-549 Cell Line or B2M overexpression A-549 Cell Line?
The choice depends on whether you are studying B2M in NSCLC context for checkpoint inhibitor resistance and lung cancer immunotherapy research. The Knockout line is the standard tool for asking whether B2M is required for these processes in NSCLC context — B2M loss-of-function is a documented mechanism of acquired resistance to pembrolizumab/nivolumab in NSCLC. Overexpression is useful for studying B2M gain-of-function effects.
For NSCLC immunotherapy research, the EDITGENE B2M Knockout in A-549 is highly relevant — A-549 is an NSCLC cell line, and B2M loss has been characterized in pembrolizumab/nivolumab-resistant NSCLC patient tumors. This product complements the parallel B2M Knockouts in HEK293T (biochemistry) and A-375 (melanoma) for cross-background validation. Rescue with WT B2M is the standard specificity control. The knockout is valuable for studying NSCLC immune evasion mechanisms and emerging immunotherapy resistance approaches.
What are the application scenarios for this model?
Primary applications:
• NSCLC checkpoint inhibitor resistance: in heterologous T cell co-culture systems, anti-PD-1/anti-PD-L1 antibody-induced T cell killing analysis — B2M KO eliminates MHC I presentation and reduces CD8+ T cell recognition.
• MHC class I-restricted antigen presentation: in NSCLC context, characterization of B2M-dependent antigen presentation.
• Cross-background validation: parallel analysis with B2M KOs in HEK293T and A-375 (both available).
EDITGENE recommends this NSCLC model for researchers investigating lung cancer immunotherapy resistance mechanisms.
Is this B2M Knockout A-549 Cell Line compatible with overexpression rescue experiments?
Yes. B2M rescue in A-549 supports NSCLC immunotherapy research:
• Construct design: same considerations as B2M/HEK293T rescue.
• NSCLC tumor antigen presentation: WT B2M rescue restores MHC I-restricted neoantigen presentation in NSCLC context.
• Pembrolizumab/nivolumab response: in heterologous tumor-T cell co-culture systems, anti-PD-1-induced tumor killing should be restored by B2M rescue.
• Functional readout: rescue should restore HLA-ABC surface expression and CD8+ T cell recognition.
A-549 transduces efficiently with lentivirus and supports stable rescue line generation.
