Description

TRIM41 (Tripartite Motif Containing 41) encodes a member of the tripartite motif (TRIM) family, characterized by a RING finger domain, B-box zinc finger, and coiled-coil region. The protein functions as an E3 ubiquitin ligase, mediating ubiquitination and subsequent proteasomal degradation of target proteins. TRIM41 is involved in innate immune signaling, particularly in the regulation of interferon responses and antiviral defense. It also plays roles in cell proliferation and differentiation.

Mutation functional classification

Loss of Function (LOF)

Frameshift and nonsense mutations that truncate the RING or B-box domains are predicted to abolish E3 ligase activity.

Gain of Function (GOF)

Missense mutations in the coiled-coil region may enhance substrate binding or alter specificity, but evidence is limited.

Dominant Negative (DN)

Mutations that disrupt dimerization or substrate recognition could interfere with wild-type TRIM41 function.

Pathways

• Ubiquitin mediated proteolysis (KEGG: hsa04120)
• RIG-I-like receptor signaling pathway (KEGG: hsa04622)
• Innate immune system (Reactome: R-HSA-168249)

Protein Summary

TRIM41 is a 574-amino acid protein with a molecular weight of approximately 65 kDa. It contains an N-terminal RING finger domain (residues 15-55) responsible for E3 ubiquitin ligase activity, two B-box zinc fingers (residues 90-130 and 140-180), a coiled-coil region (residues 200-300) mediating oligomerization, and a C-terminal domain of unknown function. The protein localizes to the cytoplasm and is involved in ubiquitination of viral proteins and cellular regulators.
