Description
FOSL1 (FOS Like 1, AP-1 Transcription Factor Subunit) encodes a leucine zipper protein that dimerizes with JUN proteins to form the AP-1 transcription factor complex. FOSL1 regulates gene expression in response to growth factors, cytokines, and stress signals, playing critical roles in cell proliferation, differentiation, apoptosis, and transformation. Overexpression of FOSL1 is frequently observed in various cancers and is associated with aggressive tumor phenotypes.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Breast Cancer | FOSL1 overexpression promotes invasion and metastasis via AP-1 target genes (e.g., MMPs, uPA) | PMID: 21502514; COSMIC |
| Lung Cancer | FOSL1 amplification and overexpression drive cell proliferation and resistance to EGFR inhibitors | PMID: 26921328; COSMIC |
| Colorectal Cancer | FOSL1 upregulation correlates with poor prognosis and activates Wnt/β-catenin signaling | PMID: 25695633; COSMIC |
| Osteosarcoma | FOSL1 is a direct target of FOS and contributes to tumorigenesis | PMID: 15988017; COSMIC |
| Inflammatory Diseases | FOSL1 modulates cytokine production in macrophages and is linked to rheumatoid arthritis | PMID: 20037588 |

Mutation functional classification
Loss of Function (LOF)
No well-characterized loss-of-function mutations reported in human disease; experimental knockout in mice shows reduced cell proliferation.
Gain of Function (GOF)
Gene amplification and overexpression in multiple cancers act as gain-of-function, enhancing AP-1 transcriptional activity and promoting oncogenic phenotypes.
Dominant Negative (DN)
No dominant-negative mutations described in FOSL1; dominant-negative effects are typically mediated by truncated FOS family members.

Protein Summary
FOSL1 (FRA-1) is a 271-amino acid protein (UniProt P15407) belonging to the FOS family of basic leucine zipper (bZIP) transcription factors. It forms heterodimers with JUN proteins to bind AP-1 sites in DNA. FOSL1 lacks a transactivation domain present in other FOS members, making it a weaker transcriptional activator. It is rapidly induced by growth factors and stress signals and is stabilized by phosphorylation. FOSL1 is overexpressed in many cancers and contributes to invasion, metastasis, and drug resistance.
