Description

SERPINE1 (serpin family E member 1) encodes plasminogen activator inhibitor-1 (PAI-1), a serine protease inhibitor that regulates fibrinolysis by inhibiting tissue-type (tPA) and urokinase-type (uPA) plasminogen activators. PAI-1 is a key modulator of blood coagulation, extracellular matrix turnover, cell adhesion, and migration. Elevated PAI-1 levels are associated with thrombotic disorders, fibrosis, and poor prognosis in various cancers. The gene is located on chromosome 7q22.1 and is primarily expressed in adipose tissue, liver, and vascular endothelium.

Protein Summary

Plasminogen activator inhibitor-1 (PAI-1) is a 45 kDa glycoprotein belonging to the serpin superfamily. It inhibits tPA and uPA by forming stable covalent complexes, thereby regulating plasmin generation and fibrinolysis. PAI-1 exists in active, latent, and substrate conformations. Its expression is induced by cytokines, growth factors, and hypoxia. Elevated PAI-1 levels are a biomarker for thrombotic risk and poor cancer prognosis. The protein is stored in platelets and secreted by endothelial cells, adipocytes, and hepatocytes.
