Description

SENP7 (SUMO specific peptidase 7) encodes a member of the sentrin/SUMO-specific protease family. The protein catalyzes the deconjugation of SUMO (small ubiquitin-like modifier) from target proteins, regulating processes such as chromatin remodeling, DNA repair, and transcriptional control. SENP7 is localized to the nucleus and is involved in the deSUMOylation of histone deacetylases and other chromatin-associated factors.

Mutation functional classification

Loss of Function (LOF)

Frameshift and nonsense variants that truncate the catalytic domain are predicted to abolish SUMO protease activity.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported in literature or databases.

Dominant Negative (DN)

Not described; dominant-negative effects have not been experimentally validated for SENP7.

Protein Summary

SENP7 is a 984-amino acid nuclear protease that specifically removes SUMO2/3 chains from target proteins. It contains a conserved C-terminal peptidase domain (C48 family) and an N-terminal regulatory region. SENP7 plays a key role in chromatin decondensation by deSUMOylating HDAC2 and other repressive complexes, thereby influencing gene expression and DNA repair. Its dysregulation is implicated in several cancers.
