Description
The PVR gene encodes CD155, a type I transmembrane glycoprotein belonging to the nectin-like family. It functions as a cell adhesion molecule and serves as the receptor for poliovirus. CD155 is involved in immune regulation through interactions with TIGIT, CD226, and CD96, and plays a role in tumor progression and viral entry.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Poliomyelitis | PVR is the cellular receptor for poliovirus; viral entry via CD155 leads to infection and neuronal damage. | UniProt, NCBI |
| Colorectal cancer | Overexpression of CD155 promotes tumor invasion and metastasis; interaction with TIGIT suppresses antitumor immunity. | COSMIC, PubMed |
| Glioma | CD155 expression correlates with malignancy; promotes cell migration and invasion. | COSMIC, PubMed |
| Hepatocellular carcinoma | Elevated CD155 expression associated with poor prognosis; modulates immune evasion. | COSMIC, PubMed |

Mutation functional classification
Loss of Function (LOF)
Rare loss-of-function mutations may impair CD155-mediated immune regulation, potentially increasing susceptibility to viral infections or altering tumor surveillance.
Gain of Function (GOF)
Gain-of-function mutations leading to overexpression or enhanced binding to TIGIT could promote immune evasion in tumors.
Dominant Negative (DN)
No dominant-negative mutations reported to date.

Protein Summary
CD155 is a 70 kDa type I transmembrane protein with three immunoglobulin-like domains in the extracellular region. It interacts with nectin and nectin-like proteins, and serves as a ligand for TIGIT, CD226, and CD96. CD155 is involved in cell adhesion, migration, and immune modulation. It is overexpressed in many cancers and contributes to immune evasion by inhibiting T cell and NK cell activity.
