Description
MMP11 (Matrix Metallopeptidase 11), also known as stromelysin-3, encodes a member of the matrix metalloproteinase (MMP) family. Unlike most MMPs, MMP11 is secreted as a proenzyme and activated extracellularly by furin. It plays a role in extracellular matrix remodeling, cell growth, and tissue repair. Overexpression is associated with various cancers and fibrotic diseases.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Breast Carcinoma | Overexpression promotes tumor invasion and metastasis via ECM degradation | PMID: 10646886 |
| Colorectal Cancer | Elevated MMP11 correlates with poor prognosis and lymph node metastasis | PMID: 19029980 |
| Lung Cancer | MMP11 expression enhances stromal remodeling and tumor progression | PMID: 21558396 |
| Osteoarthritis | MMP11 contributes to cartilage degradation in joint disease | PMID: 10700367 |

Mutation functional classification
Loss of Function (LOF)
Frameshift and nonsense mutations are predicted to cause loss of proteolytic activity.
Gain of Function (GOF)
Missense mutations in the catalytic domain may alter substrate specificity, but evidence is limited.
Dominant Negative (DN)
No dominant-negative mutations have been characterized for MMP11.

Protein Summary
MMP11 (stromelysin-3) is a 488-amino acid secreted metalloproteinase with a signal peptide, prodomain, catalytic domain containing the zinc-binding motif, and a hemopexin-like domain. It is activated by furin cleavage and degrades extracellular matrix components such as fibronectin, laminin, and collagen IV. Unlike other MMPs, it is not inhibited by TIMPs. Overexpression in tumors correlates with poor prognosis.
