Description

KLF5 (Kruppel Like Factor 5) is a member of the Kruppel-like factor (KLF) family of zinc finger transcription factors. It regulates gene expression by binding to GC-rich promoter regions. KLF5 plays critical roles in cell proliferation, differentiation, migration, and apoptosis. It is involved in embryonic development, cardiovascular homeostasis, and is frequently dysregulated in various cancers, acting as both an oncogene and tumor suppressor depending on cellular context.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast Cancer KLF5 overexpression promotes cell proliferation and migration via transcriptional activation of target genes such as FGF-BP and cyclin D1. PMID: 21804532; COSMIC
Prostate Cancer KLF5 loss or reduced expression correlates with tumor progression; acts as a tumor suppressor by regulating p53 and PTEN pathways. PMID: 18794899; COSMIC
Cardiovascular Disease KLF5 regulates vascular smooth muscle cell proliferation and phenotypic switching, contributing to atherosclerosis and restenosis. PMID: 15761150; OMIM
Colorectal Cancer KLF5 is overexpressed and promotes tumor growth through Wnt/β-catenin signaling. PMID: 20010872; COSMIC

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.1018C>T (p.Arg340Cys) Missense <1% Altered DNA-binding affinity; reported in COSMIC
c.1129G>A (p.Gly377Ser) Missense <1% Unknown functional effect; rare in ClinVar
c.1246_1247insA (p.Thr416Asnfs*5) Frameshift <1% Loss of function; truncation of C-terminal domain
Mutation functional classification

Loss of Function (LOF)

Frameshift and nonsense mutations that truncate the zinc finger domain lead to loss of DNA-binding and transcriptional activity.

Gain of Function (GOF)

Missense mutations in the activation domain may enhance transcriptional activity, though evidence is limited.

Dominant Negative (DN)

Truncated KLF5 isoforms lacking the transactivation domain can interfere with wild-type KLF5 function.

Pathways

• Wnt signaling pathway (Reactome: R-HSA-195721)
• p53 pathway (Reactome: R-HSA-3700989)
• Transcriptional regulation by KLF5 (Reactome: R-HSA-8939211)

Protein Summary

KLF5 encodes a 457-amino acid protein containing three C-terminal C2H2-type zinc fingers that mediate sequence-specific DNA binding to GC-rich elements. The N-terminal domain contains a transactivation domain. KLF5 is predominantly nuclear and functions as a transcriptional activator or repressor depending on interacting partners. It is highly expressed in the gastrointestinal tract and is implicated in cell cycle regulation, stem cell maintenance, and epithelial-mesenchymal transition. Post-translational modifications include phosphorylation and acetylation, which modulate its stability and activity.
