Description

ZNF143 (zinc finger protein 143) encodes a C2H2-type zinc finger transcription factor that binds to the SphI post-octamer element and activates small nuclear RNA (snRNA) and small nucleolar RNA (snoRNA) gene promoters. It also regulates protein-coding genes involved in cell cycle, DNA repair, and metabolism. ZNF143 is ubiquitously expressed and has been implicated in cancer progression and developmental processes.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Colorectal cancer ZNF143 overexpression promotes cell proliferation and invasion via transcriptional activation of target genes. PMID: 29774082
Breast cancer ZNF143 upregulation correlates with poor prognosis and enhances metastasis through EMT induction. PMID: 31092905
Hepatocellular carcinoma ZNF143 silencing reduces tumor growth and induces apoptosis in liver cancer cells. PMID: 31591469

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.1045C>T (p.Arg349Trp) Missense 0.02% (gnomAD) Unknown functional effect; located in zinc finger domain
c.1234G>A (p.Gly412Arg) Missense 0.01% (gnomAD) May alter DNA-binding affinity
c.789_790insA (p.Glu264Argfs*12) Frameshift <0.01% (COSMIC) Loss of function; truncation of C-terminal domain
Mutation functional classification

Loss of Function (LOF)

Frameshift and nonsense mutations in ZNF143 are predicted to cause loss of DNA-binding and transcriptional activation, potentially impairing snRNA/snoRNA expression.

Gain of Function (GOF)

Missense mutations in the zinc finger region may alter target specificity or increase transcriptional activity, but evidence is limited.

Dominant Negative (DN)

No dominant-negative mutations have been reported for ZNF143.

Pathways

• snRNA transcription by RNA polymerase II
• RNA polymerase III transcription
• Cell cycle regulation

Protein Summary

ZNF143 is a 625-amino acid protein containing seven C2H2-type zinc finger domains. It localizes to the nucleus and functions as a sequence-specific transcription factor. The protein binds to the SphI motif (GTGCC) in snRNA and snoRNA gene promoters and recruits RNA polymerase II/III complexes. ZNF143 also interacts with chromatin remodeling factors and is involved in maintaining genomic stability.
