Description
YBX1 encodes the Y-box binding protein 1, a multifunctional protein involved in transcription, translation, RNA splicing, DNA repair, and stress response. It binds to Y-box elements in promoters and to RNA, regulating gene expression at multiple levels. YBX1 is implicated in cell proliferation, differentiation, and apoptosis, and is overexpressed in many cancers, contributing to tumor progression, drug resistance, and metastasis.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Cancer (multiple types) | Overexpression of YBX1 promotes cell proliferation, invasion, and chemoresistance via regulation of genes like EGFR, MYC, and ABCB1. | High expression correlates with poor prognosis in breast, lung, colorectal, and ovarian cancers (multiple studies). |
| Prostate cancer | YBX1 is overexpressed and associated with aggressive disease; it regulates androgen receptor signaling. | Immunohistochemistry and mRNA studies show elevated YBX1 in prostate tumors. |
| Renal cell carcinoma | YBX1 expression is elevated and linked to tumor progression and metastasis. | Studies show YBX1 promotes epithelial-mesenchymal transition (EMT) in renal cancer cells. |
| Inflammatory diseases | YBX1 modulates inflammatory responses via regulation of cytokine expression. | Preclinical models suggest YBX1 involvement in inflammation, but clinical evidence is limited. |

Mutation functional classification
Loss of Function (LOF)
No clear loss-of-function mutations reported; YBX1 is essential for cell viability, and complete loss is likely lethal.
Gain of Function (GOF)
Overexpression or amplification of YBX1 is common in cancers, acting as an oncogene by enhancing cell proliferation and survival.
Dominant Negative (DN)
No dominant-negative mutations described; however, some mutations may alter protein interactions, but evidence is lacking.

Protein Summary
YBX1 is a 36 kDa protein with a cold-shock domain (CSD) that binds to Y-box sequences in DNA and RNA. It shuttles between nucleus and cytoplasm, regulating transcription, translation, and mRNA stability. Post-translational modifications (phosphorylation, ubiquitination) modulate its activity. YBX1 is overexpressed in many cancers and is a potential therapeutic target.
