Aliases WIF-1, WIF1a, WIF1b

Description

WIF1 encodes a secreted protein that binds to WNT proteins and inhibits their signaling. It acts as a tumor suppressor by antagonizing the canonical WNT/β-catenin pathway. WIF1 is frequently silenced by promoter methylation in various cancers, leading to aberrant WNT activation.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Colorectal cancer Promoter hypermethylation silences WIF1, leading to constitutive WNT signaling PMID: 17384680
Lung cancer Loss of WIF1 expression via methylation contributes to tumor progression PMID: 15696293
Prostate cancer WIF1 downregulation correlates with metastasis and poor prognosis PMID: 20010866
Osteoarthritis WIF1 polymorphisms associated with cartilage degradation PMID: 23300811

Mutation functional classification

Loss of Function (LOF)

Promoter methylation and truncating mutations reduce WIF1 expression or activity, leading to unchecked WNT signaling.

Gain of Function (GOF)

Not reported.

Dominant Negative (DN)

Not reported.

Pathways

• Wnt signaling pathway (KEGG: hsa04310)
• Negative regulation of Wnt signaling (Reactome: R-HSA-4641262)

Protein Summary

WIF1 is a 379-amino-acid secreted glycoprotein containing a WIF domain and five EGF-like domains. It binds directly to WNT ligands, preventing their interaction with Frizzled receptors and thereby inhibiting both canonical and non-canonical WNT signaling. The protein is expressed in multiple tissues and is frequently lost in cancers due to epigenetic silencing.
