Description

VPS37B (VPS37B subunit of ESCRT-I) encodes a component of the endosomal sorting complex required for transport I (ESCRT-I). This complex is essential for multivesicular body (MVB) formation, sorting of ubiquitinated membrane proteins into intraluminal vesicles, and downregulation of activated receptors. VPS37B is involved in cytokinesis, HIV-1 budding, and autophagy. Mutations or dysregulation of VPS37B have been implicated in cancer progression and neurodegenerative disorders.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Hepatocellular carcinoma Reduced VPS37B expression correlates with poor prognosis; may impair receptor downregulation PMID: 23149919
Breast cancer Low VPS37B expression associated with aggressive tumor features and shorter survival PMID: 23792583
Prostate cancer VPS37B downregulation linked to increased invasion and metastasis PMID: 25605248

Protein Summary

VPS37B is a 285-amino acid protein that forms part of the ESCRT-I complex, which also includes VPS28, VPS37A/C/D, and TSG101. It contains a mod(r) domain and a coiled-coil region. VPS37B localizes to endosomal membranes and is required for the sorting of ubiquitinated cargo into intraluminal vesicles. It also participates in abscission during cytokinesis and in the release of enveloped viruses such as HIV-1.
