Description

TUT7 (Terminal Uridylyl Transferase 7) encodes a member of the terminal uridylyltransferase (TUTase) family. The protein catalyzes the addition of uridine residues to the 3' end of RNA molecules, playing critical roles in microRNA (miRNA) processing, RNA stability, and telomere maintenance. TUT7 is involved in the uridylation of pre-let-7 miRNA, regulating its maturation, and participates in the regulation of telomerase activity. It is also implicated in the cellular response to DNA damage and viral infection.

Loss of Function (LOF)

Nonsense and frameshift mutations in TUT7 lead to truncated proteins lacking the catalytic TUTase domain, impairing RNA uridylation activity.

Gain of Function (GOF)

No gain-of-function mutations have been reported for TUT7.

Dominant Negative (DN)

No dominant-negative mutations have been described for TUT7.

• RNA surveillance (3' end uridylation)

Protein Summary

TUT7 is a 1495-amino acid protein containing a CCHC-type zinc finger domain and a terminal uridylyltransferase (TUTase) domain. It localizes to both the cytoplasm and nucleus. The enzyme adds a short poly(U) tail to the 3' end of specific RNA substrates, including pre-let-7 miRNA and telomerase RNA component (TERC). This modification influences RNA stability, processing, and function. TUT7 interacts with other RNA-binding proteins and is regulated by post-translational modifications.
