Description
TUBGCP6 encodes a component of the gamma-tubulin ring complex (γ-TuRC), which is essential for microtubule nucleation and centrosome function. The protein localizes to the centrosome and is required for proper mitotic spindle assembly and cell division. Mutations in TUBGCP6 cause autosomal recessive primary microcephaly with or without chorioretinopathy.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Primary microcephaly (MCPH) | Loss-of-function mutations impair centrosome duplication and microtubule nucleation, leading to reduced neuronal progenitor proliferation | OMIM #610053; multiple families reported |
| Microcephaly with chorioretinopathy | Disrupted centrosome function affects retinal development and brain growth | Case reports in ClinVar and literature |

Protein Summary
TUBGCP6 (GCP6) is a 1,682-amino-acid protein that serves as a structural component of the γ-tubulin ring complex (γ-TuRC). It contains a conserved GCP6 domain and zinc-binding motifs. The protein is essential for anchoring γ-tubulin at the centrosome and for efficient microtubule nucleation. Loss of TUBGCP6 function leads to centrosome abnormalities, impaired mitotic progression, and reduced brain size.
