Description

TUBGCP4 encodes a component of the gamma-tubulin ring complex (γ-TuRC), which is essential for microtubule nucleation and centrosome function. The protein localizes to the centrosome and is required for proper mitotic spindle assembly and cell division. Mutations in TUBGCP4 are associated with autosomal recessive primary microcephaly and chorioretinopathy.

Mutation functional classification

Loss of Function (LOF)

Nonsense and frameshift mutations lead to truncated protein and loss of γ-TuRC function.

Gain of Function (GOF)

No evidence of gain-of-function mutations.

Dominant Negative (DN)

Not reported; disease is autosomal recessive.

Pathways

• Microtubule nucleation (Reactome R-HSA-380259)
• Centrosome maturation (Reactome R-HSA-380270)

Protein Summary

TUBGCP4 (GCP4) is a 76 kDa component of the gamma-tubulin ring complex (γ-TuRC). It is essential for microtubule nucleation at centrosomes and spindle assembly. The protein contains a conserved GCP4 domain and interacts with other γ-TuRC subunits. Loss of function leads to centrosome abnormalities and impaired neurogenesis, underlying microcephaly.
