Description
The TTL gene encodes tubulin tyrosine ligase, an enzyme that catalyzes the post-translational addition of a tyrosine residue to the C-terminal end of detyrosinated alpha-tubulin. This reaction is part of the tubulin tyrosination/detyrosination cycle, which regulates microtubule stability, dynamics, and interactions with microtubule-associated proteins. TTL is essential for neuronal development, cell division, and ciliary function. Loss of TTL function leads to abnormal microtubule detyrosination and has been implicated in neurodegenerative disorders and cancer.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Primary microcephaly | Loss-of-function mutations in TTL impair microtubule dynamics during neurogenesis, leading to reduced brain size | OMIM #608291; PMID: 25446528 |
| Intellectual disability | Disrupted tubulin tyrosination cycle affects neuronal migration and synaptic function | ClinVar; PMID: 25446528 |
| Cancer (various) | TTL downregulation or loss promotes tumor cell invasiveness and aneuploidy via altered microtubule stability | COSMIC; PMID: 19690332 |

Mutation functional classification
Loss of Function (LOF)
Nonsense and start-loss mutations lead to complete or near-complete loss of TTL activity, causing primary microcephaly and intellectual disability.
Gain of Function (GOF)
No gain-of-function mutations have been reported for TTL.
Dominant Negative (DN)
Some missense variants (e.g., p.Arg575Trp) may act in a dominant-negative manner by interfering with dimerization or substrate binding, though evidence is limited.

Protein Summary
Tubulin tyrosine ligase (TTL) is a 42 kDa cytoplasmic enzyme that catalyzes the ATP-dependent addition of a tyrosine residue to the C-terminal glutamic acid of detyrosinated alpha-tubulin. This reaction is essential for maintaining the tyrosination/detyrosination cycle of microtubules. TTL is highly expressed in the brain and testis, and its activity is critical for proper neuronal migration, axon guidance, and cell division. Loss of TTL function leads to increased detyrosinated tubulin, which alters microtubule dynamics and has been linked to microcephaly, intellectual disability, and cancer progression.
