Description
TRIM7 (Tripartite Motif Containing 7) encodes a member of the tripartite motif (TRIM) family. The protein contains a RING finger domain, B-box, and coiled-coil region, and functions as an E3 ubiquitin ligase. TRIM7 is involved in innate immune responses, viral restriction (e.g., norovirus, SARS-CoV-2), and modulation of signaling pathways including NF-κB and mTOR. It also plays a role in cell proliferation and has been implicated in various cancers.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Colorectal cancer | TRIM7 overexpression promotes cell proliferation and invasion via ubiquitination of p53 and activation of AKT signaling. | PMID: 31525673 |
| Hepatocellular carcinoma | TRIM7 acts as a tumor suppressor by promoting degradation of the oncoprotein c-Myc. | PMID: 32047112 |
| Viral infection (Norovirus) | TRIM7 restricts norovirus replication by ubiquitinating the viral NS6/7 protease. | PMID: 33357406 |
| Viral infection (SARS-CoV-2) | TRIM7 ubiquitinates the SARS-CoV-2 envelope protein, limiting viral assembly. | PMID: 34079125 |

Mutation functional classification
Loss of Function (LOF)
Frameshift mutations (e.g., p.Thr334Asnfs*12) are predicted to cause premature truncation and loss of E3 ligase activity.
Gain of Function (GOF)
Not well characterized; overexpression in colorectal cancer suggests potential gain-of-function in oncogenic contexts.
Dominant Negative (DN)
No confirmed dominant-negative mutations reported.

Pathways

• NF-kappa B signaling pathway (KEGG: hsa04064)
• mTOR signaling pathway (Reactome: R-HSA-165159)
Protein Summary
TRIM7 is a 574-amino acid protein with a molecular weight of approximately 65 kDa. It contains an N-terminal RING finger domain (required for E3 ligase activity), two B-box domains, a coiled-coil region, and a C-terminal PRY/SPRY domain. The protein localizes to the cytoplasm and nucleus. TRIM7 mediates ubiquitination of target proteins such as p53, c-Myc, and viral proteases, thereby regulating cell cycle, apoptosis, and antiviral responses.
