Description

TRIM16 (Tripartite Motif Containing 16) encodes a member of the tripartite motif (TRIM) family. The protein contains a RING-type zinc finger, B-box, and coiled-coil domains, and functions as an E3 ubiquitin ligase. TRIM16 is involved in autophagy regulation, innate immune responses, and acts as a tumor suppressor in several cancers. It is also known as estrogen-responsive B-box protein (EBBP).

Loss of Function (LOF)

Frameshift mutations (e.g., p.Thr374Asnfs*5) are predicted to cause loss of E3 ligase activity and autophagy regulation.

Gain of Function (GOF)

No gain-of-function mutations reported in COSMIC or ClinVar.

Dominant Negative (DN)

No dominant-negative mutations reported.

Protein Summary

TRIM16 is a 574-amino acid protein with a molecular weight of approximately 65 kDa. It contains a RING finger domain (residues 25-65) that confers E3 ubiquitin ligase activity, two B-box domains, and a coiled-coil region. The protein localizes to the cytoplasm and cytosol, and is involved in ubiquitination of target proteins to regulate autophagy and innate immune signaling. TRIM16 is also implicated in the suppression of tumor growth and metastasis in multiple cancer types.
