Description
TP73 (tumor protein p73) is a member of the p53 family of transcription factors. It encodes multiple isoforms due to alternative promoter usage and splicing, including full-length TAp73 (pro-apoptotic, tumor suppressive) and N-terminally truncated DeltaNp73 (anti-apoptotic, oncogenic). TP73 regulates cell cycle arrest, apoptosis, and differentiation, and is involved in neurodevelopment, ciliogenesis, and genomic stability. Mutations and altered expression of TP73 are implicated in various cancers and developmental disorders.

Mutation functional classification
Loss of Function (LOF)
Missense and nonsense mutations in the DNA-binding domain (e.g., p.Arg273His, p.Arg278Trp, p.Arg390*) impair transactivation of target genes, reducing apoptosis and cell cycle arrest.
Gain of Function (GOF)
Overexpression of DeltaNp73 isoforms acts as a dominant-negative inhibitor of p53 and TAp73, promoting cell survival and proliferation.
Dominant Negative (DN)
DeltaNp73 isoforms lack the N-terminal transactivation domain and can heterodimerize with p53 and TAp73, blocking their transcriptional activity.

Protein Summary
TP73 encodes the tumor protein p73, a transcription factor with a DNA-binding domain, a tetramerization domain, and a C-terminal SAM domain. It shares structural homology with p53 and p63. The full-length TAp73 isoform activates pro-apoptotic and cell cycle arrest genes (e.g., BAX, CDKN1A/p21), while the DeltaNp73 isoform lacks the N-terminal transactivation domain and acts as a dominant-negative inhibitor. Post-translational modifications include phosphorylation, acetylation, and ubiquitination, regulating stability and activity. TP73 is essential for neurodevelopment, ciliogenesis, and genomic stability.
