Description

TOB1 (Transducer of ERBB2, 1) encodes a member of the TOB/BTG family of anti-proliferative proteins. The protein interacts with ERBB2 and is involved in cell cycle regulation, particularly in the G1/S transition. It also participates in TGF-beta signaling by interacting with SMAD proteins. TOB1 is considered a tumor suppressor, with reduced expression observed in various cancers.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Lung cancer Reduced TOB1 expression correlates with poor prognosis; loss of growth suppression PMID: 15696293
Breast cancer TOB1 downregulation promotes proliferation and metastasis PMID: 19680224
Colorectal cancer TOB1 inhibits Wnt signaling; decreased expression linked to tumor progression PMID: 21947006

Mutation functional classification

Loss of Function (LOF)

Frameshift and nonsense mutations that truncate the protein, impairing anti-proliferative activity.

Gain of Function (GOF)

Not reported in TOB1.

Dominant Negative (DN)

Not reported in TOB1.

Pathways

• TGF-beta signaling pathway (KEGG: hsa04350)
• ErbB signaling pathway (KEGG: hsa04012)
• Cell cycle (KEGG: hsa04110)

Protein Summary

TOB1 is a 345-amino acid protein with a conserved N-terminal domain (TOB domain) and a C-terminal region that mediates interactions with ERBB2 and SMAD proteins. It functions as a transcriptional co-repressor, inhibiting cell cycle progression by regulating cyclin D1 and c-Myc expression. The protein is ubiquitously expressed and shuttles between the nucleus and cytoplasm.
