Description

TNFSF8 (Tumor Necrosis Factor Superfamily Member 8) encodes CD30 ligand (CD30L), a type II transmembrane glycoprotein belonging to the tumor necrosis factor (TNF) family. CD30L binds to CD30 (TNFRSF8) and regulates cell proliferation, differentiation, and apoptosis, particularly in lymphoid cells. It is involved in immune responses and has been implicated in Hodgkin lymphoma, anaplastic large cell lymphoma, and other inflammatory conditions.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Hodgkin lymphoma CD30L-CD30 signaling promotes survival and proliferation of Hodgkin and Reed-Sternberg cells via NF-κB activation. ClinVar, OMIM

Protein Summary

CD30 ligand (CD30L) is a 234-amino acid type II transmembrane protein with a C-terminal extracellular TNF homology domain. It forms homotrimers and binds to CD30 (TNFRSF8) on the surface of activated T cells, B cells, and Hodgkin/Reed-Sternberg cells. CD30L-CD30 interaction triggers downstream signaling via TRAF2/5 and NF-κB, leading to cell survival, proliferation, and cytokine production. Soluble CD30L is generated by proteolytic cleavage and can modulate immune responses.
