Description
TNFSF14 (TNF Superfamily Member 14), also known as LIGHT, is a cytokine belonging to the tumor necrosis factor (TNF) ligand family. It is expressed on activated T cells, natural killer cells, and immature dendritic cells. LIGHT binds to its receptors HVEM (TNFRSF14) and LTβR, playing critical roles in co-stimulation of T cells, lymphoid organogenesis, and immune homeostasis. Dysregulation of TNFSF14 is implicated in autoimmune diseases, graft-versus-host disease, and various cancers.

Protein Summary
TNFSF14 encodes a 240-amino-acid type II transmembrane protein (LIGHT) with a C-terminal extracellular TNF homology domain. It forms homotrimers and binds to HVEM and LTβR. LIGHT is cleaved by metalloproteinases to generate a soluble form. The protein is involved in T cell co-stimulation, dendritic cell maturation, and lymphoid tissue development. Its dysregulation contributes to inflammatory and malignant diseases.
