Description

TNFRSF9 (CD137, 4-1BB) encodes a member of the tumor necrosis factor (TNF) receptor superfamily. This transmembrane protein is expressed on activated T cells, NK cells, dendritic cells, and other immune cells. It provides co-stimulatory signals that promote T cell proliferation, survival, and cytokine production. TNFRSF9 is a key target in cancer immunotherapy, with agonist antibodies (e.g., urelumab, utomilumab) under investigation. The gene is also implicated in autoimmune diseases and graft-versus-host disease.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Lymphoma (various subtypes) Overexpression of TNFRSF9 on tumor-infiltrating lymphocytes may contribute to immune evasion; somatic mutations in the gene have been reported in B-cell lymphomas. COSMIC; ClinVar
Autoimmune diseases (e.g., rheumatoid arthritis, systemic lupus erythematosus) Increased soluble CD137 levels and altered TNFRSF9 signaling promote chronic inflammation and autoantibody production. OMIM; PubMed
Graft-versus-host disease (GVHD) TNFRSF9 activation on donor T cells exacerbates GVHD; blockade or genetic deletion reduces severity in models. PubMed

Hotspot Mutations
Variant Type Frequency Functional Description
c.487G>A (p.Gly163Arg) Missense <0.1% (rare) Unknown; reported in COSMIC for lymphoid neoplasms

Mutation functional classification

Loss of Function (LOF)

Rare missense or nonsense mutations that impair receptor expression or ligand binding, leading to reduced T cell co-stimulation.

Gain of Function (GOF)

Not well characterized; some somatic mutations in lymphoma may enhance signaling, but evidence is limited.

Dominant Negative (DN)

No confirmed dominant-negative mutations reported in TNFRSF9.

Gene Ontology (GO)

• GO:0005031 – tumor necrosis factor-activated receptor activity • GO:0007165 – signal transduction
• GO:0042102 – positive regulation of T cell proliferation • GO:0006915 – apoptotic process
• GO:0006955 – immune response

Protein Summary

TNFRSF9 (CD137, 4-1BB) is a 255-amino-acid type I transmembrane glycoprotein with four cysteine-rich extracellular domains characteristic of the TNFR superfamily. It forms homodimers and binds to its ligand TNFSF9 (4-1BBL). Upon ligand binding, it recruits TRAF adaptor proteins (TRAF1, TRAF2, TRAF3) and activates NF-κB, JNK, and PI3K/Akt signaling pathways, leading to enhanced T cell survival, proliferation, and effector function. The protein is a target for cancer immunotherapy (agonist antibodies) and for modulating autoimmune inflammation.
