Description

TLE3 encodes a member of the TLE (transducin-like enhancer of split) family of transcriptional corepressors. These proteins are homologs of the Drosophila Groucho protein and function by interacting with transcription factors such as TCF/LEF to repress Wnt target gene expression. TLE3 is involved in cell fate determination, differentiation, and cancer progression.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Colorectal cancer TLE3 represses Wnt/β-catenin target genes; loss of TLE3 expression may enhance Wnt signaling and tumorigenesis PMID: 19029980
Breast cancer TLE3 overexpression correlates with poor prognosis and may modulate estrogen receptor signaling PMID: 23382212
Ovarian cancer TLE3 expression is associated with chemoresistance and epithelial-mesenchymal transition PMID: 25695676

Mutation functional classification

Loss of Function (LOF)

Nonsense and frameshift mutations in TLE3 are predicted to cause loss of corepressor activity, potentially derepressing Wnt target genes.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported in TLE3.

Dominant Negative (DN)

No dominant-negative mutations described for TLE3.

Pathways

• Wnt signaling pathway (KEGG: hsa04310)
• Transcriptional misregulation in cancer (KEGG: hsa05202)
• Hippo signaling pathway (KEGG: hsa04390)

Protein Summary

TLE3 is a 781-amino-acid nuclear protein containing an N-terminal glutamine-rich domain and a C-terminal WD40 repeat domain. It functions as a transcriptional corepressor by recruiting histone deacetylases to promoter regions. TLE3 interacts with TCF/LEF transcription factors to repress Wnt/β-catenin target genes, and with other factors such as RUNX1 and PAX5. The protein is widely expressed in adult tissues and plays roles in neurogenesis, hematopoiesis, and cancer.
