Description

THY1 encodes a glycosylphosphatidylinositol (GPI)-anchored cell surface glycoprotein, also known as CD90. It is a member of the immunoglobulin superfamily and plays roles in cell adhesion, neurite outgrowth, T-cell activation, and tumor suppression. THY1 is expressed on thymocytes, neurons, endothelial cells, and fibroblasts, and its expression is frequently lost in various cancers.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (multiple types) Loss of THY1 expression promotes epithelial-mesenchymal transition and metastasis. NCBI Gene, COSMIC
Pulmonary fibrosis Reduced THY1 on lung fibroblasts correlates with myofibroblast differentiation and fibrosis. NCBI Gene, PubMed
Glomerulosclerosis THY1 depletion in mesangial cells leads to proliferative glomerulonephritis in animal models. OMIM, PubMed
Alzheimer's disease THY1 interacts with amyloid-beta and modulates neurite degeneration. UniProt, PubMed

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.1A>G (p.Met1?) Missense <0.1% Likely loss of start codon; functional impact unknown
c.215C>T (p.Pro72Leu) Missense <0.1% Rare variant; no known disease association
Mutation functional classification

Loss of Function (LOF)

Loss of THY1 expression is common in cancers and fibrosis, leading to increased cell migration and proliferation.

Gain of Function (GOF)

Not reported; THY1 overexpression is generally tumor-suppressive.

Dominant Negative (DN)

Not described for THY1.

Protein Summary

Thy-1 (CD90) is a 25-37 kDa GPI-anchored glycoprotein with a single immunoglobulin-like domain. It is expressed on thymocytes, neurons, endothelial cells, and fibroblasts. Thy-1 mediates cell-cell and cell-matrix adhesion, modulates integrin signaling, and regulates T-cell activation. Loss of Thy-1 is associated with cancer progression and fibrosis.
