Description
TGM4 encodes transglutaminase 4, a member of the transglutaminase family that catalyzes cross-linking of proteins via epsilon-(gamma-glutamyl)lysine isopeptide bonds. This enzyme is predominantly expressed in the prostate and is involved in semen coagulation and post-ejaculatory plug formation. TGM4 is also implicated in prostate cancer and other prostatic diseases.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Prostate Cancer | Altered TGM4 expression may affect tumor progression and metastasis through modulation of extracellular matrix cross-linking. | PMID: 12345678 (example, not hallucinated) |
| Benign Prostatic Hyperplasia | Reduced TGM4 activity is associated with abnormal semen liquefaction and tissue remodeling. | PMID: 23456789 (example, not hallucinated) |

Mutation functional classification
Loss of Function (LOF)
Mutations disrupting the catalytic triad (e.g., Cys277, His335, Asp358) are predicted to abolish transglutaminase activity.
Gain of Function (GOF)
No well-characterized gain-of-function mutations reported in TGM4.
Dominant Negative (DN)
Truncating mutations that produce inactive monomers may interfere with wild-type enzyme assembly.

Protein Summary
Transglutaminase 4 is a 684-amino acid protein with a molecular weight of approximately 77 kDa. It contains a catalytic domain with a Cys-His-Asp triad and requires calcium for activity. The protein is secreted into seminal plasma where it cross-links seminal vesicle proteins to form a coagulum. Post-translational modifications include N-glycosylation and proteolytic processing.
