Description

TFDP2 (Transcription factor Dp-2) encodes a member of the DP family of transcription factors that heterodimerize with E2F proteins to regulate genes involved in cell cycle progression, DNA replication, and apoptosis. The protein forms complexes with E2F1–E2F6, modulating transcriptional activation or repression depending on the cellular context. TFDP2 is ubiquitously expressed and plays a critical role in development and tumorigenesis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (various types) Dysregulation of E2F/DP complexes leads to uncontrolled cell proliferation; TFDP2 overexpression or amplification observed in some tumors. COSMIC, ClinVar
Developmental disorders Altered TFDP2 expression may disrupt normal cell cycle control during embryogenesis. OMIM, NCBI Gene

Mutation functional classification

Loss of Function (LOF)

Nonsense or frameshift mutations that truncate the protein, impairing dimerization with E2F and transcriptional activity.

Gain of Function (GOF)

Amplification or overexpression leading to increased E2F target gene activation and enhanced cell proliferation.

Dominant Negative (DN)

Mutations that produce a truncated DP2 protein capable of binding E2F but unable to activate transcription, thereby inhibiting normal E2F/DP function.

Protein Summary

The TFDP2 protein (UniProt Q14188) is 410 amino acids long and contains a conserved DP domain required for heterodimerization with E2F transcription factors. It localizes to the nucleus and regulates expression of genes controlling G1/S transition. Post-translational modifications include phosphorylation, which modulates its activity and stability.
