Description
TFDP1 (Transcription Factor Dp-1) encodes a member of the DP family of transcription factors. The protein forms heterodimers with E2F family members, thereby regulating the expression of genes involved in cell cycle progression, DNA replication, and apoptosis. TFDP1 is essential for the transcriptional activity of E2F complexes and is a key component of the retinoblastoma (RB) tumor suppressor pathway. Alterations in TFDP1 expression or function have been implicated in various cancers.

Mutation functional classification
Loss of Function (LOF)
Frameshift and nonsense mutations that truncate the protein, impairing dimerization with E2F and transcriptional activation.
Gain of Function (GOF)
Gene amplification or overexpression leading to increased E2F activity and enhanced cell cycle progression.
Dominant Negative (DN)
Missense mutations in the dimerization domain that disrupt E2F binding but retain partial function, potentially interfering with wild-type DP1.

Protein Summary
TFDP1 encodes the 410-amino acid Transcription Factor Dp-1 protein. It contains a conserved DNA-binding/dimerization domain that mediates heterodimerization with E2F transcription factors. The DP1-E2F complex binds to E2F-responsive promoters to regulate genes essential for G1/S transition. Post-translational modifications, including phosphorylation, modulate its activity. TFDP1 is ubiquitously expressed with highest levels in testis and lymphoid tissues. Its dysregulation is linked to multiple cancer types.
