Description

TFCP2 (Transcription Factor CP2) encodes a member of the CP2 family of transcription factors. The protein binds to the alpha-globin promoter and regulates erythroid gene expression. It also functions in cell cycle control, differentiation, and has been implicated in viral transcription (e.g., HIV-1 LTR). TFCP2 is involved in both transcriptional activation and repression, and its dysregulation is associated with several cancers.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Hepatocellular carcinoma TFCP2 overexpression promotes cell proliferation and metastasis via transcriptional activation of target genes. COSMIC; PMID: 25619955
Colorectal cancer TFCP2 upregulation correlates with poor prognosis and enhances Wnt/β-catenin signaling. COSMIC; PMID: 28341829
Prostate cancer TFCP2 acts as an oncogene by regulating androgen receptor signaling and cell cycle genes. COSMIC; PMID: 23382210
Breast cancer TFCP2 overexpression associated with aggressive tumor phenotypes and chemoresistance. COSMIC; PMID: 25944712

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.1015C>T (p.Arg339Trp) Missense <0.1% Altered DNA-binding affinity; reported in COSMIC
c.1246G>A (p.Glu416Lys) Missense <0.1% Unknown functional impact; rare population variant
c.1480_1481insA (p.Thr494Asnfs*12) Frameshift <0.1% Predicted loss of function; truncation
Mutation functional classification

Loss of Function (LOF)

Frameshift and nonsense mutations (e.g., p.Thr494Asnfs*12) are predicted to cause loss of function via protein truncation.

Gain of Function (GOF)

Overexpression and amplification of TFCP2 in multiple cancers suggest gain-of-function oncogenic activity, though specific activating point mutations are not well characterized.

Dominant Negative (DN)

No dominant-negative mutations have been reported for TFCP2 in curated databases.

Pathways

• Erythropoiesis (alpha-globin gene regulation)
• Wnt/β-catenin signaling (in cancer)
• Androgen receptor signaling (prostate cancer)

Protein Summary

TFCP2 (also known as LSF) is a 502-amino acid transcription factor containing a CP2-type DNA-binding domain. It forms homo- or heterodimers with other CP2 family members (e.g., TFCP2L1) and binds to the consensus sequence CNRGN(N)5CNRGN. The protein regulates genes involved in erythroid differentiation, cell cycle progression, and apoptosis. Post-translational modifications include phosphorylation and sumoylation, which modulate its activity. In cancer, TFCP2 is frequently overexpressed and promotes tumor growth, invasion, and metastasis.
