Description

TEAD4 encodes a member of the transcriptional enhancer factor (TEF) family, which binds to the MCAT element in muscle-specific genes. It is a key downstream effector of the Hippo signaling pathway, forming complexes with YAP1 and WWTR1 (TAZ) to regulate cell proliferation, differentiation, and organ size. TEAD4 is essential for embryonic development, particularly in trophectoderm specification, and is implicated in various cancers.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer TEAD4 overexpression promotes YAP1-mediated transcription, driving proliferation and metastasis. PMID: 25977310
Colorectal cancer TEAD4 amplification and activation via Hippo pathway mutations enhance tumor growth. PMID: 24930764
Gastric cancer TEAD4 upregulation correlates with poor prognosis and activates Wnt/β-catenin signaling. PMID: 28411376
Hepatocellular carcinoma TEAD4 overexpression promotes invasion and EMT via YAP1 interaction. PMID: 26638074

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.101C>T (p.Pro34Leu) Missense <0.1% Unknown functional impact
c.458G>A (p.Arg153Gln) Missense <0.1% Reported in COSMIC; potential loss of DNA binding

Mutation functional classification

Loss of Function (LOF)

Mutations disrupting DNA-binding domain (e.g., p.Arg153Gln) reduce transcriptional activity.

Gain of Function (GOF)

Mutations enhancing YAP1 binding or nuclear localization (e.g., p.Arg238Trp) increase oncogenic signaling.

Dominant Negative (DN)

Truncating mutations that interfere with wild-type TEAD4 function are rare.

Pathways

Hippo signaling pathway (KEGG: hsa04390)
Transcriptional regulation by the AP-2 family (Reactome: R-HSA-8864260)
Signaling by Hippo (Reactome: R-HSA-2028269)

Protein Summary

TEAD4 is a 434-amino acid transcription factor containing a TEA/ATTS DNA-binding domain and a C-terminal YAP1-binding domain. It forms a complex with YAP1/TAZ to activate target genes involved in cell growth and survival. Post-translational modifications include phosphorylation and acetylation, which modulate its activity. TEAD4 is widely expressed in adult tissues, with highest levels in heart, skeletal muscle, and placenta.
