Description
TEAD3 (TEA Domain Transcription Factor 3) is a member of the TEAD family of transcription factors, which share a conserved TEA/ATTS DNA-binding domain. TEAD3 binds to the MCAT element (5'-CATTCCT-3') in the promoters of target genes and acts as a nuclear effector of the Hippo signaling pathway by interacting with YAP1 and WWTR1/TAZ coactivators. It plays critical roles in embryonic development, cell proliferation, and organ size control. TEAD3 is expressed in multiple tissues, including placenta, heart, and skeletal muscle.
Disease Associations

|---|---|---|
| Cancer (various types) | TEAD3 overexpression or aberrant activation via Hippo pathway dysregulation promotes cell proliferation and tumorigenesis | COSMIC, ClinVar |
| Developmental disorders | TEAD3 mutations may disrupt Hippo signaling, affecting organogenesis and growth | OMIM |
| Cardiovascular disease | TEAD3 regulates cardiac gene expression; altered activity linked to heart development defects | NCBI Gene, UniProt |

Mutation functional classification
Loss of Function (LOF)
Frameshift and nonsense mutations that truncate the protein, impairing DNA binding or YAP/TAZ interaction, are classified as loss-of-function.
Gain of Function (GOF)
Missense mutations that enhance TEAD3 stability or coactivator binding may act as gain-of-function, but specific examples are not well documented.
Dominant Negative (DN)
Mutations that retain DNA binding but fail to activate transcription could act as dominant-negative, though no confirmed cases are reported.

Protein Summary
TEAD3 encodes a 435-amino acid protein containing an N-terminal TEA DNA-binding domain and a C-terminal transactivation domain that interacts with YAP1 and WWTR1/TAZ. The protein functions as a transcription factor that regulates genes involved in cell proliferation, differentiation, and organ size control. TEAD3 is ubiquitously expressed with highest levels in placenta, heart, and skeletal muscle. Post-translational modifications include phosphorylation and acetylation, which modulate its activity. TEAD3 is implicated in cancer and developmental disorders through dysregulation of Hippo signaling.
