Description
TEAD2 encodes a member of the TEA domain family of transcription factors, which bind to the MCAT element in gene promoters. It is a key downstream effector of the Hippo signaling pathway, regulating cell proliferation, differentiation, and organ size. TEAD2 interacts with YAP1 and WWTR1 (TAZ) to activate target genes involved in development and cancer.

Mutations & Variants
Hotspot Mutations
| Variant | Type | Frequency | Functional Description |
|---|---|---|---|
| c.119C>T (p.Pro40Leu) | Missense | Rare | Alters DNA-binding affinity; associated with Sveinsson chorioretinal atrophy |
| c.457G>A (p.Glu153Lys) | Missense | Rare | Potential loss of function; reported in COSMIC |
| Amplification | Copy number gain | Frequent in cancers | Increases TEAD2 expression; activates Hippo target genes |
Mutation functional classification
Loss of Function (LOF)
Missense variants in the DNA-binding domain (e.g., p.Pro40Leu) reduce transcriptional activity.
Gain of Function (GOF)
Gene amplification or overexpression leads to enhanced YAP/TAZ-mediated transcription.
Dominant Negative (DN)
Not well characterized; some missense variants may interfere with wild-type TEAD2 function.

Protein Summary
TEAD2 is a 445-amino acid transcription factor containing a TEA DNA-binding domain and a YAP/TAZ-binding domain. It forms complexes with YAP1 or WWTR1 to regulate genes involved in cell growth, apoptosis, and development. Post-translational modifications include phosphorylation and acetylation, modulating its activity.
