Description

TBX2 (T-box transcription factor 2) is a member of the T-box family of transcription factors, characterized by a conserved DNA-binding domain (T-box). It plays critical roles in embryonic development, particularly in heart, limb, and neural crest cell patterning. TBX2 acts as a transcriptional repressor by binding to T-box elements in target gene promoters. It is involved in cell cycle regulation, senescence suppression, and apoptosis inhibition. Aberrant expression or mutation of TBX2 is associated with several cancers, including breast, lung, and melanoma, as well as developmental disorders such as ulnar-mammary syndrome and congenital heart defects.

Mutation functional classification

Loss of Function (LOF)

Missense and nonsense mutations (e.g., p.Pro34Leu, p.Arg412*) reduce or abolish DNA binding and transcriptional repression, leading to developmental defects such as ulnar-mammary syndrome.

Gain of Function (GOF)

Gene amplification and overexpression in cancers (e.g., breast, lung) enhance TBX2-mediated repression of p21 and p19ARF, promoting cell proliferation and inhibiting senescence.

Dominant Negative (DN)

Some missense mutations (e.g., p.Gly153Arg) may produce a protein that interferes with wild-type TBX2 function, though dominant-negative effects are not well-characterized for TBX2.

Pathways

• T-box transcription factor pathway (Reactome: R-HSA-5619507)
• p53-independent G1/S DNA damage checkpoint (KEGG: hsa04110)
• Senescence and autophagy in cancer (WikiPathways: WP615)

Protein Summary

TBX2 is a 712-amino acid transcription factor containing a conserved T-box DNA-binding domain. It functions primarily as a transcriptional repressor, binding to T-box elements (TBE) in target gene promoters. Key targets include CDKN1A (p21), CDKN2A (p19ARF), and NPPA. TBX2 represses p21 and p19ARF, thereby inhibiting cell cycle arrest and senescence. The protein localizes to the nucleus and interacts with other transcription factors such as GATA4 and NKX2-5. Post-translational modifications include phosphorylation, which may modulate its activity. TBX2 is essential for heart development, limb patterning, and neural crest cell migration. In cancer, TBX2 overexpression contributes to tumorigenesis by bypassing senescence and promoting proliferation.
