Description

The TBCE gene encodes tubulin folding cofactor E, a key chaperone protein involved in the post-translational folding of α-tubulin and the assembly of microtubules. TBCE is part of a multimeric tubulin-folding complex that includes cofactors A–E. It specifically binds to and stabilizes the native conformation of α-tubulin, enabling its heterodimerization with β-tubulin. Mutations in TBCE cause autosomal recessive disorders such as hypoparathyroidism, retardation, and dysmorphism (HRD) syndrome and Kenny-Caffey syndrome type 1 (KCS1), characterized by skeletal abnormalities, growth retardation, and immune dysfunction.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Hypoparathyroidism, retardation, and dysmorphism syndrome (HRD) Loss-of-function mutations in TBCE impair α-tubulin folding, disrupting microtubule dynamics and cellular processes in parathyroid, skeletal, and neuronal tissues. OMIM #241410; ClinVar
Kenny-Caffey syndrome type 1 (KCS1) Biallelic TBCE mutations lead to defective microtubule assembly, causing cortical thickening of long bones, short stature, and transient hypocalcemia. OMIM #244460; ClinVar
Sanjad-Sakati syndrome Same as HRD syndrome; caused by homozygous TBCE mutations in Middle Eastern populations. OMIM #241410; ClinVar

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.155-166del (p.Val52_Gly55del) Deletion Found in HRD/KCS1 patients Loss of function; disrupts tubulin binding
c.250C>T (p.Arg84*) Nonsense Rare Premature truncation; loss of function
c.434G>A (p.Arg145Gln) Missense Reported in KCS1 Impaired α-tubulin folding activity
Mutation functional classification

Loss of Function (LOF)

Most TBCE mutations are loss-of-function, leading to reduced or absent cofactor E activity, defective α-tubulin folding, and microtubule instability.

Gain of Function (GOF)

No gain-of-function mutations have been reported for TBCE.

Dominant Negative (DN)

No dominant-negative effects have been described; TBCE disorders are autosomal recessive.

Pathways

• Tubulin folding and microtubule assembly (Reactome: R-HSA-389977)
• Formation of tubulin folding intermediates (Reactome: R-HSA-389958)

Protein Summary

Tubulin folding cofactor E (TBCE) is a 527-amino-acid cytosolic protein that acts as a chaperone in the α-tubulin folding pathway. It binds to quasi-native α-tubulin released from the chaperonin TRiC/CCT, stabilizing it and facilitating its heterodimerization with β-tubulin. TBCE contains a CAP-Gly domain essential for microtubule plus-end binding and a leucine-rich repeat (LRR) domain for protein-protein interactions. Defects in TBCE cause severe developmental syndromes due to impaired microtubule function.
