Description

TADA2A encodes a component of the SAGA (Spt-Ada-Gcn5-acetyltransferase) and ATAC (Ada-Two-A-containing) histone acetyltransferase complexes. It functions as a transcriptional adaptor that bridges sequence-specific transcription factors to the general transcription machinery and modulates chromatin acetylation, thereby regulating gene expression. TADA2A is involved in development, cell cycle control, and DNA repair.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Neurodevelopmental disorder with intellectual disability and dysmorphic features Loss-of-function mutations in TADA2A impair SAGA complex activity, leading to aberrant gene expression during neurodevelopment ClinVar, OMIM
Breast cancer Somatic mutations and altered expression of TADA2A may disrupt transcriptional regulation of tumor suppressor genes COSMIC, NCBI
Colorectal cancer Recurrent missense mutations in TADA2A observed in tumor samples; potential role in chromatin remodeling COSMIC

Mutation functional classification

Loss of Function (LOF)

Nonsense, frameshift, and start-loss variants that truncate or abolish TADA2A protein, leading to reduced SAGA complex activity.

Gain of Function (GOF)

Missense mutations in the acidic domain may enhance interaction with transcription factors, observed in some cancers.

Dominant Negative (DN)

Missense mutations in the SAGA interaction domain that disrupt complex formation while retaining partial function.

Pathways

• Chromatin modifying enzymes (Reactome: R-HSA-3244102)
• Transcriptional regulation by the SAGA complex (Reactome: R-HSA-3214847)
• Gene expression (Reactome: R-HSA-74160)

Protein Summary

TADA2A is a 443-amino-acid protein (UniProt O75478) containing a SANT domain and an ADA2 homology region. It interacts with GCN5 (KAT2A) and TADA3 to form the catalytic core of the SAGA complex. TADA2A stabilizes the complex and enhances histone H3 acetylation. It is ubiquitously expressed with highest levels in testis and brain. Post-translational modifications include phosphorylation and acetylation.
