Description
TADA1 (Transcriptional Adaptor 1) encodes a core subunit of the ATAC (Ada Two A-containing) histone acetyltransferase complex. The protein directly interacts with GCN5 (KAT2A) and is essential for complex integrity and histone H3 acetylation. TADA1 functions in transcriptional coactivation, chromatin remodeling, and cell cycle regulation. It is ubiquitously expressed with highest levels in testis and lymphoid tissues.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Cancer (pan-cancer) | Altered expression of TADA1 may disrupt ATAC complex activity, affecting chromatin accessibility and oncogene/tumor suppressor gene transcription. | COSMIC; TCGA |
| Neurodevelopmental disorders | Rare missense variants in TADA1 have been reported in patients with intellectual disability and developmental delay, suggesting a role in neuronal gene regulation. | ClinVar; Decipher |

Mutation functional classification
Loss of Function (LOF)
Frameshift and nonsense mutations that truncate the protein or cause nonsense-mediated decay are classified as loss-of-function. These are rare and may contribute to developmental phenotypes.
Gain of Function (GOF)
No gain-of-function mutations have been reported for TADA1.
Dominant Negative (DN)
No dominant-negative mutations have been characterized for TADA1.

Protein Summary
TADA1 is a 42 kDa nuclear protein containing a conserved N-terminal domain that mediates interaction with GCN5 and other ATAC subunits. It does not possess intrinsic enzymatic activity but is required for the structural integrity and histone acetyltransferase activity of the ATAC complex. The protein is involved in acetylation of histone H3 at lysines 9 and 14, facilitating open chromatin and gene activation. TADA1 also interacts with transcription factors such as p53 and MYC, linking it to cell proliferation and stress responses.
