Description
TACR2 encodes the tachykinin receptor 2 (NK2R), a G protein-coupled receptor that binds neurokinin A (NKA) with high affinity. Activation of NK2R stimulates phospholipase C, leading to intracellular calcium mobilization and smooth muscle contraction. The receptor is primarily expressed in peripheral tissues such as the respiratory tract, gastrointestinal tract, and urinary bladder, where it mediates bronchoconstriction, gut motility, and nociception. TACR2 is a therapeutic target for asthma, irritable bowel syndrome, and chronic pain.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Asthma | NK2R activation in airway smooth muscle induces bronchoconstriction and mucus secretion. | ClinVar, OMIM |
| Irritable Bowel Syndrome (IBS) | Increased NK2R signaling in colonic smooth muscle contributes to hypermotility and visceral hypersensitivity. | NCBI Gene, PubMed |
| Chronic Pain | NK2R mediates nociceptive transmission in peripheral and central pain pathways. | OMIM, UniProt |
| Urinary Incontinence | NK2R activation in detrusor muscle promotes bladder contraction. | NCBI Gene |

Protein Summary
The tachykinin receptor 2 (NK2R) is a 398-amino acid integral membrane protein with seven transmembrane domains. It belongs to the rhodopsin-like GPCR family. Upon binding neurokinin A, NK2R couples primarily to Gq/11 proteins, activating phospholipase C and increasing intracellular calcium. The receptor is expressed in smooth muscle cells of the respiratory, gastrointestinal, and urogenital tracts, and in certain neurons. NK2R antagonists are under investigation for asthma, IBS, and pain management.
