Description

SUN2 encodes a nuclear envelope protein that is a component of the LINC (Linker of Nucleoskeleton and Cytoskeleton) complex. It spans the inner nuclear membrane and interacts with nesprins in the perinuclear space, connecting the nuclear lamina to the cytoskeleton. SUN2 is involved in nuclear positioning, cell migration, mechanotransduction, and DNA repair. Dysregulation of SUN2 has been implicated in cancer progression, muscular dystrophies, and laminopathies.

Protein Summary

SUN2 is a 717-amino acid protein (UniProt Q9UH99) with an N-terminal nucleoplasmic domain, a transmembrane domain, and a C-terminal SUN domain that localizes to the perinuclear space. It forms homotrimers and interacts with nesprin-2 (SYNE2) and lamin A/C (LMNA). SUN2 is essential for tethering the nucleus to the cytoskeleton, regulating nuclear movement, and maintaining nuclear shape. Post-translational modifications include phosphorylation at Ser48 and Ser52, which modulate its interaction with partners.
