Description
The SUMO2 gene encodes a member of the small ubiquitin-like modifier (SUMO) protein family. SUMO2 is conjugated to target proteins in a process called SUMOylation, a post-translational modification that regulates protein function, localization, and stability. SUMO2 shares high sequence similarity with SUMO3 and can form poly-SUMO chains, which are critical for cellular stress responses, DNA damage repair, and cell cycle control. SUMO2 is ubiquitously expressed and plays a key role in maintaining genome integrity and cellular homeostasis.

Mutation functional classification
Loss of Function (LOF)
Loss-of-function mutations in SUMO2 are rare and may impair SUMOylation capacity, leading to defects in DNA repair and cell cycle regulation, potentially contributing to genomic instability.
Gain of Function (GOF)
Gain-of-function mutations that increase SUMO2 expression or enhance its conjugation activity could promote oncogenic signaling by stabilizing tumor-promoting proteins.
Dominant Negative (DN)
Dominant-negative mutations might interfere with SUMO2 incorporation into poly-SUMO chains, disrupting normal SUMOylation dynamics and downstream signaling.

Protein Summary
SUMO2 is a 95-amino acid protein that is post-translationally processed to its mature form. It is conjugated to target proteins via an isopeptide bond, often forming poly-SUMO chains. SUMO2 shares ~97% sequence identity with SUMO3 but differs from SUMO1. SUMO2 is involved in stress-induced SUMOylation, where it rapidly conjugates to many nuclear proteins upon heat shock or oxidative stress. It plays critical roles in transcriptional regulation, DNA repair, and maintenance of genome stability. Dysregulation of SUMO2 has been linked to various diseases, including cancer and neurodegeneration.
