Description
STX3 encodes syntaxin 3, a member of the syntaxin family of SNARE proteins. Syntaxin 3 is primarily involved in the docking and fusion of vesicles with target membranes, playing a critical role in exocytosis and membrane trafficking in epithelial cells, neurons, and immune cells. Mutations in STX3 cause microvillus inclusion disease type 2 (MVID2), a severe congenital enteropathy.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Microvillus inclusion disease 2 (MVID2) | Loss-of-function mutations disrupt apical membrane trafficking in enterocytes, leading to microvillus atrophy and intractable diarrhea. | ClinVar, OMIM |
| Congenital diarrhea | Impaired SNARE-mediated vesicle fusion results in defective nutrient absorption. | NCBI Gene, OMIM |

Protein Summary
Syntaxin 3 is a 301-amino acid protein with a C-terminal transmembrane domain and a SNARE motif that mediates interactions with other SNARE proteins. It localizes to the plasma membrane and is essential for regulated exocytosis in polarized epithelial cells, particularly in the intestine. The protein forms a complex with SNAP23 and VAMP8 to facilitate vesicle fusion.
