Description

STX12 encodes syntaxin 12, a member of the syntaxin family of t-SNARE proteins. It localizes to early endosomes and the trans-Golgi network, where it mediates vesicle docking and fusion, playing a critical role in endosomal recycling, autophagy, and synaptic vesicle recycling. STX12 is ubiquitously expressed with high levels in the brain and is essential for neuronal development and function.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Neurodevelopmental disorder with intellectual disability and speech delay Biallelic loss-of-function variants impair endosomal trafficking, disrupting synaptic function and neuronal connectivity. ClinVar (Pathogenic variants reported)
Schizophrenia (susceptibility) Altered STX12 expression may affect glutamatergic signaling and synaptic plasticity. OMIM (association studies)
Cancer (various types) Dysregulation of STX12 affects membrane trafficking, potentially influencing tumor invasion and metastasis. COSMIC (somatic mutations in multiple cancers)

Mutation functional classification

Loss of Function (LOF)

Biallelic loss-of-function variants (nonsense, frameshift, splice) cause neurodevelopmental disorder, likely due to haploinsufficiency or complete loss of STX12 function.

Gain of Function (GOF)

No evidence of gain-of-function mutations; somatic missense variants in cancer are not characterized as activating.

Dominant Negative (DN)

Not reported; no dominant-negative mechanism established.

Protein Summary

Syntaxin 12 is a 276-amino acid protein with a C-terminal transmembrane domain and a SNARE coiled-coil domain. It forms a complex with VAMP4 and SNAP-47 to mediate fusion of early endosomes and recycling vesicles. STX12 is critical for neurite outgrowth, synaptic plasticity, and autophagosome maturation. Its dysfunction is linked to neurodevelopmental disorders and cancer progression.
