Description

STMN4 (stathmin 4) encodes a member of the stathmin family of microtubule-destabilizing phosphoproteins. The protein, also known as RB3, regulates microtubule dynamics by sequestering tubulin heterodimers and promoting catastrophe. STMN4 is predominantly expressed in the nervous system and plays roles in neuronal differentiation, axon guidance, and synaptic plasticity. Its expression is altered in various cancers and neurological disorders.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer Overexpression of STMN4 promotes cell proliferation and migration via microtubule destabilization and enhanced mitotic spindle dynamics. PMID: 25605274
Lung adenocarcinoma Upregulation of STMN4 correlates with poor prognosis and increased metastatic potential through regulation of epithelial-mesenchymal transition. PMID: 29367642
Neuroblastoma STMN4 is highly expressed in aggressive neuroblastoma subtypes and contributes to chemoresistance by modulating microtubule stability. PMID: 23575477
Alzheimer's disease Altered STMN4 expression in hippocampal neurons may affect synaptic integrity and microtubule-associated transport. PMID: 20628086

Protein Summary

STMN4 (stathmin 4) is a 150-amino acid protein (UniProt Q9H169) belonging to the stathmin family. It contains a conserved stathmin-like domain that mediates tubulin binding and microtubule destabilization. The protein is regulated by phosphorylation at multiple serine residues (e.g., Ser16, Ser63) by kinases such as CDK1 and PKA, which modulate its activity during the cell cycle and neuronal differentiation. STMN4 is predominantly cytoplasmic and enriched in growth cones and synaptic regions.
