Description
STEAP2 (STEAP family member 2) encodes a six-transmembrane protein that functions as a metalloreductase, reducing Fe3+ to Fe2+ and Cu2+ to Cu1+, thereby facilitating metal ion uptake. It is highly expressed in prostate tissue and is implicated in prostate cancer progression, iron homeostasis, and oxidative stress response. STEAP2 is also involved in cell proliferation and may play a role in tumorigenesis.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Prostate Cancer | Overexpression of STEAP2 promotes cell proliferation and invasion; may regulate androgen receptor signaling. | COSMIC: increased expression in prostate tumors; PMID: 14559847 |
| Colorectal Cancer | Upregulation of STEAP2 correlates with poor prognosis; may enhance iron uptake to support tumor growth. | COSMIC: somatic mutations; PMID: 25689618 |
| Iron Metabolism Disorders | Altered STEAP2 expression affects iron homeostasis, potentially contributing to anemia or iron overload. | UniProt: function in iron reduction; PMID: 16959783 |

Mutation functional classification
Loss of Function (LOF)
Frameshift and nonsense mutations that truncate the protein lead to loss of metalloreductase activity, impairing iron and copper uptake.
Gain of Function (GOF)
Missense mutations that increase enzyme activity or protein stability could enhance metal uptake and promote tumor growth, though not well-documented.
Dominant Negative (DN)
No evidence for dominant-negative effects; STEAP2 likely functions as a homodimer, but mutations that disrupt dimerization could exert dominant-negative effects, yet not reported.

Protein Summary
STEAP2 is a 490-amino acid transmembrane protein with six membrane-spanning domains. It localizes to the plasma membrane and endosomes, where it reduces Fe3+ and Cu2+ using NADPH as an electron donor. The protein forms homodimers and may interact with other STEAP family members. STEAP2 is highly expressed in the prostate and is upregulated in prostate cancer, where it promotes cell growth and survival. It also plays a role in iron homeostasis in various tissues.
