Chromosomal Location 5q35.2
NCBI Gene ID 8614 ncbi.nlm.nih.gov/gene/8614
Ensembl ID ENSG00000113739
UniProt ID O76061
OMIM ID 603665

Aliases STC-2, stanniocalcin-2

Description

STC2 encodes stanniocalcin 2, a secreted homodimeric glycoprotein that regulates calcium and phosphate homeostasis. It is induced by hypoxia via HIF-1α and modulates cell proliferation, migration, and survival. STC2 is overexpressed in multiple cancers and is associated with poor prognosis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer STC2 overexpression promotes tumor growth and metastasis via PI3K/AKT signaling PMID: 25642768
Colorectal cancer Elevated STC2 correlates with advanced stage and poor survival PMID: 27840973
Ovarian cancer STC2 enhances chemoresistance through ERK pathway activation PMID: 29351287
Hypoxia-related disorders STC2 is a direct HIF-1 target, upregulated under low oxygen PMID: 15297311

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.331C>T (p.Arg111Cys) Missense <0.1% Unknown functional impact
c.487G>A (p.Gly163Ser) Missense <0.1% Reported in COSMIC

Mutation functional classification

Loss of Function (LOF)

Frameshift insertions (e.g., c.614_615insA) likely produce truncated, non-functional protein.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported in STC2.

Dominant Negative (DN)

No evidence of dominant-negative effects for STC2 mutations.

Pathways

• HIF-1 signaling pathway (Reactome: R-HSA-1234174)
• Calcium signaling pathway (KEGG: hsa04020)
• PI3K-Akt signaling pathway (KEGG: hsa04151)

Protein Summary

Stanniocalcin 2 is a 302-amino-acid secreted glycoprotein that forms homodimers. It contains a conserved stanniocalcin domain and is involved in calcium/phosphate regulation. The protein is induced by hypoxia and can activate MAPK/ERK and PI3K/AKT signaling. It is implicated in cancer progression, chemoresistance, and metabolic disorders.
