Description

STAM2 (Signal Transducing Adaptor Molecule 2) encodes a member of the STAM family of adaptor proteins. It is a component of the ESCRT-0 complex involved in endosomal sorting of ubiquitinated cargo, including activated receptor tyrosine kinases. STAM2 also participates in cytokine and growth factor signaling pathways, regulating receptor downregulation and intracellular trafficking.

Disease Associations

Protein Summary

STAM2 is a 525-amino acid adaptor protein containing a VHS domain, a UIM (ubiquitin-interacting motif), and an SH3 domain. It forms a complex with HRS (HGS) to constitute the ESCRT-0 complex, which recognizes ubiquitinated cargo on endosomal membranes and facilitates sorting into multivesicular bodies for lysosomal degradation. STAM2 also interacts with JAK kinases and STAT transcription factors, modulating cytokine receptor signaling. Its expression is ubiquitous, with higher levels in testis and brain.
