Description
STAB1 encodes stabilin-1, a large transmembrane scavenger receptor expressed on sinusoidal endothelial cells and alternatively activated macrophages. It mediates endocytosis of modified low-density lipoproteins, advanced glycation end-products, and apoptotic cells. Stabilin-1 also functions in angiogenesis, leukocyte trafficking, and immune regulation. Its expression is upregulated in tumor-associated macrophages and inflamed tissues.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Hepatocellular carcinoma | STAB1 overexpression in tumor-associated macrophages promotes angiogenesis and immune evasion. | PMID: 25636840 |
| Colorectal cancer | High STAB1 expression correlates with poor prognosis and increased metastasis. | PMID: 30348673 |
| Atherosclerosis | Stabilin-1 mediates uptake of oxidized LDL in macrophages, contributing to foam cell formation. | PMID: 19229032 |
| Rheumatoid arthritis | STAB1 expressed on synovial macrophages facilitates leukocyte adhesion and inflammation. | PMID: 21187405 |

Mutation functional classification
Loss of Function (LOF)
Frameshift mutations (e.g., p.Leu1523fs) are predicted to truncate the protein, impairing scavenger receptor activity.
Gain of Function (GOF)
No gain-of-function mutations have been reported in STAB1.
Dominant Negative (DN)
No dominant-negative mutations have been described for STAB1.

Protein Summary
Stabilin-1 is a 2570-amino acid type I transmembrane glycoprotein with multiple fasciclin, EGF-like, and laminin-type EGF domains. It functions as a scavenger receptor for modified lipoproteins, advanced glycation end-products, and apoptotic cells. The protein is predominantly expressed on sinusoidal endothelial cells of the liver, spleen, and lymph nodes, as well as on alternatively activated macrophages. Stabilin-1 mediates cell adhesion, angiogenesis, and immune modulation, and its overexpression is linked to tumor progression and chronic inflammation.
